Hemoglobin examination is a crucial parameter in evaluating hematological conditions and the cardiovascular system. The accuracy of results highly depends on the procedures and methods used to ensure correct disease diagnosis. Objective: This study aims to determine the comparison of hemoglobin levels using the Point of Care Testing (POCT) method with a spectrophotometer at the Cendrawasih Palu Polytechnic. Materials and Methods: This type of research is descriptive with a cross-sectional study design. The research sample consisted of 10 people, with data collection conducted on January 28, 2026. Data analysis used statistical tests to observe significant differences between the two methods. Results: The examination results showed that the average hemoglobin levels with the POCT method tended to be higher than the spectrophotometry (Cyanmethemoglobin) method. Conclusions: Based on statistical tests, a P-Value > 0.05 was obtained, indicating that there was no statistically significant difference between the two methods, and the average results were within the normal range.
